About the Coronavirus testing solutions 

Testing for 2019-nCoV in humans can be briefly divided into three separate stages:

These include a robust 4X qPCR master mix for high sample volume input of up to 10 µl in 20 µl reactions. The new kits are available with different ROX concentrations and detect virus RNA in very diluted samples as of <5 RNA copies per reaction. 4X 1Step RT qPCR Probe Kits have been tested for Sars-CoV-2 detection. Our classical 1Step RT qPCR kits for a single step one tube RT qPCR from RNA templates include qPCR master mix and a blend of thermostable HighScriber™ Reverse Transcriptase with RNase Inhibitor. We know you know how it works and how to optimize your reactions… Nevertheless, take a minute to check our best PCR optimization tips for faster results in our OEM customer’s hands:

1. Tip number one - MIXING of all reagents is very important for your qPCR results

Do not underestimate the importance of mixing of all the reagents before use. It is especially important when working with bulk quantities and concentrated solutions. Highly concentrated dNTPs or buffers used without thorough mixing provide inconsistency in results due to concentration fluctuations.

2. Tip number two – optimization of CYCLING conditions is a key

Every reagents supplier suggests running the annealing temperature gradient for your first qPCR experiment with a new master mix or a new primer-probe-template set. Please, always do it.

Different buffers’ salt concentrations (ionic strengths) change the annealing behavior of your primers and probes. Each new qPCR master mix requires cycling adjustments.

highQu qPCR master mixes are of high ionic strengths and require higher annealing temperatures for the most optimal performance.

Start your experiment with highQu qPCR mixes by applying annealing gradient with 0,5-degree increments in a range of 58 ºC to 65 ºC. The cycling times may also differ from mix to mix. Do not perform too long denaturation, elongation or annealing steps, follow supplier’s recommendations first.
